TITLE: GENERAL PRINCIPALS AND GUIDELINES


Preface:

In contrast to micro-organisms dedifferentiated plant cell cultures are no complete organisms but derivatives of organisms which might not necessarily be identical with the original organisms where they have been derived from. Therefore in contrast to micro-organisms, principals for identification and authentication of cell lines cannot rely on taxonomical methods and principals but has to develop an own unique strategy for these purposes.

Presently the availability of methods for the long term preservation for plant cell cultures is limited. Therefore in many cases the maintenance of cell lines in the living state is unavoidable.

Plant cell cultures may be infected with unidentified micro-organisms. To avoid any risks for laboratory personnel in these cases safety regulations for the work with micro-organisms have to be followed.

 

Guidelines and Regulations:

Although plant cell cultures cannot be compared to micro-organisms in all terms the organisation of the plant cell culture collection should folllow, if possible, the principals and spirit laid down in the "Guidelines for the Establishment and Operation of Collections of Cultures of Micro-organisms" published by the World Federation of Culture Collections (WFCC) in and its 2nd edition of 1999 1990 (PC/1999/1/2 Appendix 1).

This manual is part of the efforts to reach a comparable and defined standard of quality among European service collections within the CABRI project. Management and organisation of the collection therefore has to follow the basic principals and objectives of the CABRI Project.

Although many dedifferentiated plant cell cultures cannot be regenerated to intact plants and often are of no use for practical plant breeding, the management and organisation of the plant cell culture collection has to be aware of the principals, guidelines and the international action plan agreed upon at the 4. International Technical Conference on Plant Genetic Resources, Leipzig 17.-23.6.1996 (in: "Schriften zu Genetischen Ressourcen - 4.
